Overview
PIK-201 (also known as PIKTOR) is an orally bioavailable small molecule inhibitor that dual-targets the PI3Kα and mTORC1/mTORC2 pathways. Developed by Faeth Therapeutics (in collaboration with Sensei Biotherapeutics), PIK-201 is designed to overcome resistance mechanisms often seen with single-node inhibitors of the PI3K/AKT/mTOR pathway. By simultaneously inhibiting PI3Kα and both mTOR complexes, the drug aims to more effectively suppress oncogenic signaling in tumors with PIK3CA mutations or PTEN loss. It is currently being evaluated in Phase 2 clinical trials for the treatment of endometrial cancer, often in combination with specific dietary interventions (metabolic therapy) to manage insulin-mediated feedback loops that can otherwise limit the efficacy of PI3K inhibitors.
