PIK-75 + ocHDL NP is a nanoparticle drug delivery system that encapsulates the small-molecule multikinase inhibitor PIK-75 (also known as F7) within organic core high-density lipoprotein-like nanoparticles (ocHDL NPs). Developed by researchers at Northwestern University Feinberg School of Medicine and City of Hope, this bio-inspired platform mimics the size, surface charge, and composition of natural human HDL, including the use of apolipoprotein A-I. The nanoparticles are designed to actively target the Scavenger Receptor Class B Type 1 (SR-B1), which is overexpressed in various malignancies, facilitating selective intracellular delivery of the hydrophobic PIK-75 cargo. Once delivered, PIK-75 acts as a potent inhibitor of p110α (PI3K), DNA-dependent protein kinase (DNA-PK), and p38γ. Preclinical studies have shown significant anti-tumor activity in models of prostate cancer, cutaneous T-cell lymphoma, acute myeloid leukemia, and pancreatic cancer.
