PIK-85 (also referred to as PIK085) is a potent and selective small-molecule inhibitor of phosphatidylinositol-3-phosphate 5-kinase (PIKfyve), a lipid kinase essential for lysosomal maturation and autophagic flux. Developed by PIKSci Inc., PIK-85 is being investigated for the treatment of multiple myeloma (MM), a malignancy characterized by high basal requirements for autophagy to maintain plasma cell homeostasis and immunoglobulin synthesis. By inhibiting PIKfyve, PIK-85 disrupts autophagic flux, induces cellular vacuolation, and triggers apoptosis in human myeloma cell lines. Preclinical studies have demonstrated that PIK-85 possesses superior potency and improved pharmacokinetic properties compared to earlier compounds like PIK001. Furthermore, PIKfyve inhibition by PIK-85 has been shown to upregulate MHC Class I expression on the surface of myeloma cells, suggesting a potential role in enhancing tumor immunogenicity and synergy with immunotherapies.
