Overview

PIK-III is a potent and highly selective small molecule inhibitor of **vacuolar protein sorting 34 (Vps34)**, which is the only member of the **Class III phosphatidylinositol 3-kinase (PI3K)** family. Developed as a chemical probe, PIK-III specifically targets the ATP-binding pocket of Vps34, blocking the synthesis of phosphatidylinositol 3-phosphate (PI3P). This lipid is a crucial mediator of **autophagy** initiation and endosomal trafficking. By inhibiting Vps34, PIK-III prevents the formation of autophagosomes and disrupts autophagic flux. In the context of oncology research, particularly for **FLT3-ITD acute myeloid leukemia (AML)**, PIK-III has demonstrated the ability to induce apoptosis in leukemic cells and reduce their repopulating capacity, especially when used in combination with mobilization agents or tyrosine kinase inhibitors. It is widely used in preclinical studies to investigate the therapeutic potential of autophagy inhibition in cancer and other diseases.
