Drug intelligence / Profile preview
PIK7 is a proteolytically stable peptide inhibitor of myosin light chain kinase (MLCK) designed to prevent endothelial hyperpermeability and vascular leakage. Developed by the National Medical Research Center of Cardiology (named after Academician E.I. Chazov) in Russia, PIK7 is based on the structure of MLC11-19, a competitive MLCK inhibitor, but modified to improve stability against plasma peptidases. In preclinical models, PIK7 has demonstrated antiedematous effects by suppressing vascular hyperpermeability caused by myocardial ischemia-reperfusion injury, suggesting potential as an adjuvant therapy for myocardial infarction. Additionally, PIK7 has been shown to inhibit platelet adhesion to fibrinogen- and collagen-coated surfaces under high blood flow conditions, indicating potential antiatherothrombotic applications.
