Overview

pIL15EAM is an optimized DNA plasmid construct encoding human interleukin-15 (IL-15), developed by the University of Pennsylvania as a molecular adjuvant for HIV immunotherapy. It is designed to be co-administered with DNA vaccines, such as PENNVAX-B, to enhance cellular immune responses by promoting the proliferation and survival of CD8+ T cells and natural killer (NK) cells. The construct has been engineered for high expression levels, significantly exceeding those of native IL-15 DNA sequences. In clinical trials, it has been evaluated for its safety and immunogenicity in HIV-infected individuals on stable antiretroviral therapy, aiming to boost the body's ability to control the virus. The plasmid is typically formulated with bupivacaine to facilitate DNA uptake into cells upon intramuscular administration.
