A dual-agent, small-molecule combination of the pan-PI3K inhibitor pilaralisib (SAR245408, XL147) and the dual PI3K/mTOR inhibitor voxtalisib (SAR245409, XL765) investigated in oncology to inhibit the PI3K/AKT/mTOR signaling pathway. Sanofi developed both agents in collaboration with Exelixis; they were evaluated individually and in combinations with endocrine therapy such as letrozole in hormone receptor–positive, HER2‑negative breast cancer and other cancers. Development of SAR245408 or SAR245409 in combination with letrozole for HR+ advanced or metastatic breast cancer was terminated before completing phase 2 enrollment. The agents were also studied in an open-label extension to assess long‑term safety when used as monotherapy or in combination regimens.[1][3][6][8][10]
