PIMA-PEG-CDDP is a polymer-drug conjugate nanoparticle consisting of a poly(isobutylene-alt-maleic acid) (PIMA) backbone derivatized with polyethylene glycol (PEG) sidechains and complexed with cisplatin (CDDP). Developed by researchers at Brigham and Women's Hospital, this nanoplatinate is designed to self-assemble into nanoparticles that improve the therapeutic index of cisplatin. By utilizing a mixture of di- and mono-carboxylato bonds and coordinate bonds to link cisplatin to the polymer, PIMA-PEG-CDDP allows for sustained release of the active platinum drug. In preclinical models of lung and breast cancer, it demonstrated enhanced therapeutic efficacy and reduced systemic and nephrotoxicity compared to free cisplatin.
