Pimecrolimus is a topical immunosuppressant of the calcineurin inhibitor class, primarily used to treat mild to moderate atopic dermatitis (eczema) in patients who cannot use or have not responded to other therapies. It works by binding with high affinity to macrophilin-12 (FKBP-12), inhibiting the calcium-dependent phosphatase calcineurin, and thereby blocking T cell activation and cytokine transcription. This results in reduced synthesis and release of inflammatory cytokines from T cells and mast cells, suppressing immune responses that drive eczema symptoms. Pimecrolimus is available as a cream for topical application[1][2][3][5][6].
