pimicotinib

Pimicotinib is an orally available, highly potent and selective small molecule inhibitor of the colony-stimulating factor 1 receptor (CSF1R; also known as CD115 or M-CSFR). Developed by Abbisko Therapeutics, it blocks CSF1R activation and downstream signaling, thereby inhibiting the activity of tumor-associated macrophages (TAMs) and myeloid-derived suppressor cells (MDSCs), reducing immune suppression in the tumor microenvironment. This mechanism enhances antitumor T-cell responses and inhibits tumor cell proliferation. Pimicotinib is primarily under investigation for tenosynovial giant cell tumor (TGCT) not amenable to surgery, with ongoing phase III trials. It has also entered clinical development for chronic graft-versus-host disease (cGVHD) and pancreatic cancer[1][3][5][7][8].
