Overview

Pimicotinib (ABSK021) is a potent, highly selective, small-molecule inhibitor of the colony-stimulating factor 1 receptor (CSF-1R) developed by Abbisko Therapeutics for the treatment of tenosynovial giant cell tumor (TGCT). Rifampicin is a macrocyclic antibiotic that also functions as a potent inducer of the cytochrome P450 3A4 (CYP3A4) enzyme. The combination of pimicotinib and rifampicin is being evaluated in a Phase 1, open-label, fixed-sequence clinical pharmacology study to determine the impact of CYP3A4 induction on the pharmacokinetics of pimicotinib. This study is essential for establishing dosing recommendations for patients who may require concomitant administration of pimicotinib and strong CYP3A4 inducers. While pimicotinib has received approval in China for TGCT, this specific combination is utilized primarily in the context of drug-drug interaction (DDI) research.
