Overview

PIN-5018 is an orally bioavailable, selective molecular glue degrader (MGD) targeting Casein kinase 1 alpha (CK1α). Developed by Pin Therapeutics, it is designed to induce the targeted proteasomal degradation of CK1α by facilitating its recruitment to the cereblon (CRBN) E3 ubiquitin ligase complex. CK1α is a serine/threonine kinase that plays a critical role in oncogenic signaling pathways, including Wnt/β-catenin and p53, which regulate cell proliferation and survival. PIN-5018 is currently being evaluated in a Phase 1 clinical trial for patients with advanced, unresectable, or metastatic solid tumors.
