Pin1 siRNA is an experimental RNA interference (RNAi) modality designed to silence the expression of the PIN1 gene, which encodes the peptidyl-prolyl cis/trans isomerase NIMA-interacting 1 protein. PIN1 is a unique prolyl isomerase that specifically binds and isomerizes phosphorylated serine/threonine–proline motifs, thereby altering the conformation and function of numerous oncogenic and tumor-suppressive proteins. Overexpression of PIN1 is common in various human cancers, including hepatocellular carcinoma, pancreatic cancer, prostate cancer, and glioblastoma, where it promotes cell cycle progression and proliferation. In preclinical research, Pin1 siRNA has been delivered using various systems such as nano-liposomes, GalNAc-conjugated nanoparticles, and thermosensitive hydrogels to inhibit tumor growth and enhance sensitivity to treatments like radiation. It is primarily used as a research tool and has not yet transitioned into a single branded clinical therapeutic asset.
