Pinatuzumab vedotin + rituximab is an investigational combination therapy evaluated primarily in relapsed or refractory B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma (DLBCL) and indolent non-Hodgkin lymphoma. Pinatuzumab vedotin is an antibody-drug conjugate (ADC) targeting CD22 on B cells; it consists of a humanized monoclonal anti-CD22 antibody linked via a protease-cleavable linker to the cytotoxic agent monomethyl auristatin E (MMAE), a microtubule-disrupting agent. Rituximab is a monoclonal antibody targeting CD20 on B cells, promoting cell lysis primarily through complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity. The combination is designed to provide synergistic targeting of malignant B cells by addressing two separate B-cell antigens. In clinical studies, this combination has demonstrated clinical activity and an acceptable safety profile in relapsed/refractory DLBCL and iNHL, but not in chronic lymphocytic leukemia (CLL)[1][2][4]. Development of the combination by Roche/Genentech has proceeded to at least phase II trials, but further commercial development and approval have not been reported.
