pING-hHER3FL is an investigational plasmid DNA cancer vaccine designed to encode the full-length human epidermal growth factor receptor type-3 (HER-3; also known as HER3 or ErbB-3). Once administered, the circular DNA fragment is taken up by host cells and leads to expression of the HER3 protein, stimulating an immune response against tumor cells expressing HER3. The primary mechanism involves antagonism of the receptor tyrosine-protein kinase erbB-3 (HER3), aiming to induce a targeted immune attack on cancers overexpressing this antigen. The drug is being developed as an immunotherapeutic agent for solid tumors with high HER3 expression—including breast, lung, gastric, head and neck, ovarian cancers, melanoma, and colorectal cancer. As of 2025 it is in Phase 1 clinical trials at Duke University[1][2][4][5][6][7].
