Overview

Pinitol (3-O-methyl-D-chiro-inositol) is a naturally occurring cyclic sugar alcohol and a methyl ether of D-chiro-inositol, primarily found in soy and various legumes. It functions as an insulin sensitizer and has been investigated for the treatment of type 2 diabetes and metabolic syndrome to improve glycemic control and insulin resistance. In the field of neurodegeneration, Humanetics Pharmaceuticals is developing a proprietary formulation of pinitol, designated NIC5-15, for Alzheimer's disease. NIC5-15 is reported to act as a Notch-sparing γ-secretase modulator, which reduces the production of amyloid-beta (Aβ) peptides while preserving the cleavage of other γ-secretase substrates like Notch. The compound has reached Phase 2 clinical trials for Alzheimer's disease and is also commercially available as a dietary supplement.
