Piperphentonamine (GZ-001) is a novel small molecule therapeutic developed by Guangzhou Zhongwei Biotechnology for the treatment of heart failure, particularly acute heart failure. As a chemical derivative of the non-selective alpha-adrenergic antagonist phentolamine, it is designed to improve hemodynamic performance through a dual mechanism of action. It functions as a vasodilator by antagonizing alpha-1 and alpha-2 adrenergic receptors, thereby reducing systemic vascular resistance and cardiac afterload. Additionally, piperphentonamine possesses positive inotropic properties that enhance myocardial contractility, distinguishing it from traditional alpha-blockers. This combined effect aims to increase cardiac output and alleviate symptoms of congestion in patients with acute decompensated heart failure without the excessive tachycardia often associated with pure vasodilators.
