Overview

This is a combination regimen of three chemotherapeutic agents—pirarubicin, mitomycin, and lobaplatin—used primarily in the treatment of cancers such as primary liver cancer and potentially other solid tumors. - **Pirarubicin** is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and RNA synthesis. - **Mitomycin** is an alkylating agent that crosslinks DNA, thereby inhibiting DNA synthesis and function. - **Lobaplatin** is a third-generation platinum-based compound that forms DNA adducts, causing apoptosis by interfering with DNA replication and transcription. The combination has been studied in transcatheter arterial chemoembolization (TACE) for primary liver cancer, where it has shown improved efficacy compared to single-agent regimens or other combinations[5][3]. The use of drug-eluting beads loaded with pirarubicin combined with lobaplatin can maintain high local concentrations at the tumor site while reducing systemic toxicity[5]. Mitomycin may be added for its synergistic cytotoxic effects.
