pIRF7 GPC3-targeted CAR T cells are an investigational cell therapy designed to treat Glypican-3 (GPC3) expressing solid tumors, specifically hepatocellular carcinoma (HCC) and hepatoblastoma (HBL). This next-generation CAR T-cell product incorporates a constitutively active (phosphorylated) form of Interferon Regulatory Factor 7 (pIRF7) to enhance antitumor activity. IRF7 was identified as a key transcriptional driver associated with therapeutic response in patients treated with earlier iterations of GPC3-CAR T cells. The addition of pIRF7 is intended to promote a more favorable T-cell phenotype (higher naïve and central memory populations), increase expansion, and improve killing efficacy and persistence within the immunosuppressive tumor microenvironment of solid tumors.
