Overview

Pirfazimine is a novel small molecule antituberculosis drug developed by the Institute of Materia Medica of the Chinese Academy of Medical Sciences. It is derived from clofazimine, retaining its core structure for efficacy against *Mycobacterium tuberculosis* while incorporating modifications to enhance pharmacokinetic properties and mitigate side effects such as skin staining. The drug has demonstrated potent activity against both drug-sensitive and drug-resistant strains of tuberculosis in preclinical studies. Additionally, pirfazimine exhibits a minimal impact on liver drug-metabolizing enzymes, making it a suitable candidate for combination therapy in the treatment of drug-resistant tuberculosis.
