Overview

Piroxantrone is an anthrapyrazole antineoplastic agent structurally related to mitoxantrone and doxorubicin. It was developed as a potential chemotherapeutic drug for the treatment of various cancers, including solid tumors and hematologic malignancies. Like other anthracenedione derivatives, piroxantrone acts primarily by intercalating into DNA and inhibiting topoisomerase II, leading to DNA strand breaks and inhibition of cell proliferation. However, clinical development of piroxantrone was discontinued due to limited efficacy and concerns about toxicity compared with other available agents.
