Overview

Pirtobrutinib + R-CHOP is an investigational combination regimen consisting of the next-generation, non-covalent (reversible) Bruton's tyrosine kinase (BTK) inhibitor pirtobrutinib and the standard-of-care immunochemotherapy regimen R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone). This combination is primarily being evaluated for the first-line treatment of newly diagnosed non-germinal center B-cell (non-GCB) diffuse large B-cell lymphoma (DLBCL). Pirtobrutinib, developed by Eli Lilly, inhibits BTK, a critical node in the B-cell receptor signaling pathway that drives the survival and proliferation of malignant B cells, particularly in the non-GCB/ABC subtype of DLBCL. By combining a reversible BTK inhibitor with the cytotoxic and immunotherapeutic components of R-CHOP, the regimen aims to overcome constitutive BCR signaling and improve clinical outcomes compared to standard immunochemotherapy alone.
