Overview

Pirtobrutinib + BEAM is an investigational combination regimen being studied as a preconditioning treatment for autologous stem cell transplantation (ASCT) in patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL). The regimen combines pirtobrutinib (Jaypirca), a highly selective, non-covalent (reversible) Bruton's tyrosine kinase (BTK) inhibitor, with the standard high-dose chemotherapy regimen BEAM, which consists of carmustine, etoposide, cytarabine, and melphalan. Pirtobrutinib is designed to inhibit both wild-type and C481-mutant BTK, providing a therapeutic option for patients who have progressed on covalent BTK inhibitors. The BEAM components function as potent cytotoxic agents to achieve deep cytoreduction before stem cell rescue. This combination is currently being evaluated in Phase 2 clinical trials sponsored by Ruijin Hospital to improve transplant outcomes and progression-free survival in aggressive B-cell lymphomas.
