Overview

Pitofenone is a small molecule antispasmodic (spasmolytic) drug used to relieve pain and spasms of smooth muscles in the stomach, intestines, urinary tract, biliary tract, and uterus. It is commonly indicated for conditions such as menstrual cramps, gastrointestinal colic, kidney stones, gallstones, and painful menstruation. Pitofenone works by suppressing muscle spasms through anticholinergic (antimuscarinic) activity—blocking acetylcholine at muscarinic receptors—and by inhibiting acetylcholinesterase. This results in relaxation of vascular and extravascular smooth muscles similar to papaverine. It is often used in combination with fenpiverinium bromide and metamizole sodium for enhanced analgesic and anti-inflammatory effects[1][2][3][4][5][6][7].
