Overview

Pitrakinra is a 15-kDa recombinant human protein based on wild-type interleukin-4 (IL-4), engineered to act as an antagonist of the interleukin-4 receptor alpha chain (IL-4Rα). By binding to IL-4Rα, pitrakinra blocks the signaling of both IL-4 and IL-13—key cytokines involved in Th2-mediated inflammation. This dual antagonism interrupts the inflammatory cascade responsible for asthma and atopic dermatitis. Pitrakinra was developed primarily for asthma and atopic eczema; it has been evaluated in multiple phase II clinical trials via subcutaneous injection and inhalation routes. The drug demonstrated anti-inflammatory effects with a favorable safety profile but development was discontinued after phase II[1][2][3][8][10].
