Overview

Pivarubicin (AD 445) is a small molecule anthracycline antitumor agent developed by Paradox Pharmaceuticals. It was designed as a hydrolytically stable alternative to benzarubicin (AD 198) to circumvent multiple mechanisms of drug resistance and provide a non-cardiotoxic profile at therapeutic doses. Pivarubicin acts as a protein kinase C (PKC) modulator, specifically binding to the C1b regulatory domain of PKC isoforms and activating PKC-delta. This activation triggers rapid mitochondrial-dependent apoptosis in cancer cells. The compound is part of Paradox Pharmaceuticals' pipeline for treating veterinary cancers, specifically canine mammary cancer, and is also under development for human triple-negative breast cancer. The tertiary trimethyl arrangement (pivalate) provides chemical stability by sterically hindering esterase-mediated cleavage while maintaining PKC-mediated activity.
