Overview

Pivekimab sunirine is a **CD123-targeting antibody-drug conjugate** developed by ImmunoGen and now owned by AbbVie. It consists of a high-affinity anti-CD123 monoclonal antibody linked via a cleavable linker to an indolinobenzodiazepine pseudodimer payload designed to alkylate DNA and induce single-strand breaks with limited cross-linking. The drug is being developed primarily for **blastic plasmacytoid dendritic cell neoplasm** and **acute myeloid leukemia**, particularly CD123-positive disease. Public sources identify it as a first-in-class ADC, with published phase 1/2 AML data, registered clinical trials in BPDCN and AML, and AbbVie later reporting a U.S. BLA submission for BPDCN in September 2025.
