PIXY321 is a recombinant fusion protein that combines human granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-3 (IL-3). It was developed to harness the complementary hematopoietic effects of both cytokines, with GM-CSF supporting myeloid progenitor cells and IL-3 promoting erythroid, megakaryocytic, and multipotential progenitor cells. PIXY321 binds to cell lines expressing receptors for either ligand and exhibits enhanced biological activity compared to individual or equimolar combinations of the two cytokines. In preclinical studies, PIXY321 accelerated neutrophil and platelet recovery in animal models subjected to sublethal irradiation. Clinically, it has been investigated for use in patients undergoing high-dose therapy (HDT) with autologous bone marrow transplantation (ABMT), as well as for mobilization of peripheral-blood cells in patients with malignant lymphoma or bone marrow defects. The drug demonstrated the ability to stimulate multilineage hematopoiesis in vivo, enhance engraftment after transplantation, and promote rapid hematopoietic recovery following chemotherapy or BMT[2][3][4][5][6].
