Drug intelligence / Profile preview
PJ008 (also known as PL5) is a PEGylated recombinant phenylalanine ammonia-lyase (PAL) being developed by Chongqing Peg-Bio Biopharm (Chongqing Paijin Biotechnology) for the treatment of phenylketonuria (PKU). Developed using the company's proprietary MAS-PEG platform technology, PJ008 features mutations in the T-cell and B-cell epitope regions of phenylalanine ammonia-lyase to reduce immunogenicity while retaining enzymatic activity. It is further modified with site-specific PEGylation to extend its half-life, enabling a potential once-weekly or once-every-two-weeks subcutaneous dosing regimen. PJ008 acts by metabolizing phenylalanine in the blood, thereby reducing toxic phenylalanine accumulation in patients with PKU who lack functional phenylalanine hydroxylase (PAH). PJ008 is currently in Phase I clinical development in China.
