Drug intelligence / Profile preview
PK-188 is a novel small-molecule allosteric, non-competitive inhibitor targeting New Delhi metallo-β-lactamase-1 (NDM-1) and Verona integron-encoded metallo-β-lactamase (VIM). It is being developed as an adjunctive agent to restore susceptibility to carbapenem antibiotics, such as meropenem, in multidrug-resistant Gram-negative bacteria, particularly carbapenem-resistant *Klebsiella pneumoniae* (CR-Kp) co-producing CTX-M and NDM-1. In preclinical studies, PK-188 demonstrated favorable safety, high stability in serum and liver microsomes, and high bioavailability when administered intraperitoneally. When combined with meropenem in a neutropenic murine pulmonary infection model, PK-188 significantly reduced pulmonary bacterial burden and restored meropenem efficacy.
