Overview

PKF115-584 binds to and inhibits the β-catenin/TCF transcriptional complex, leading to downregulation of Wnt target genes such as c-MYC and Cyclin D1, suppression of cell proliferation, cell cycle arrest, and induction of apoptosis in tumor cells that depend on Wnt signaling. In various preclinical models, PKF115-584 reduces tumor growth and increases survival, showing activity against multiple myeloma and adrenocortical carcinoma cell lines, including those resistant to other therapies. The compound does not exhibit cytotoxicity in normal cells not reliant on active Wnt signaling, suggesting potential therapeutic selectivity[1][3][4][7].
