Overview

PKM2 siRNA is a small interfering RNA designed to silence the expression of Pyruvate Kinase M2 (PKM2), an isoenzyme of pyruvate kinase that is frequently overexpressed in cancer cells and plays a critical role in the Warburg effect (aerobic glycolysis). By targeting PKM2 mRNA for degradation via the RNA interference (RNAi) pathway, this siRNA reduces PKM2 protein levels, leading to decreased metabolic activity, reduced ATP and glutathione levels, and the activation of AMP-activated protein kinase (AMPK). In preclinical studies, particularly in glioblastoma and glioma stem cells, PKM2 knockdown has been shown to inhibit cell growth and invasion while inducing apoptosis, with minimal toxicity observed in normal astrocytes.
