PKMYT1 siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of the PKMYT1 (Protein Kinase, Membrane Associated Tyrosine/Threonine 1) gene. PKMYT1 is a critical regulator of the cell cycle, specifically the G2/M transition, where it phosphorylates and inhibits CDK1. In research led by the National Cancer Institute (NCI), PKMYT1 siRNA is being evaluated as part of a synthetic lethal strategy for the treatment of Diffuse Pleural Mesothelioma (DPM). When used in combination with WEE1 siRNA, it induces robust knockdown of its target, leading to significant G2/M arrest, depletion of S-phase cells, and synergistic induction of apoptosis. The therapeutic is delivered via a specialized nanoparticle-hydrogel system designed for localized RNA delivery to anatomically restricted, treatment-refractory cancers.
