Overview

**PKNPs** are an experimental, target-responsive supramolecular nanoparticle system developed for **Alzheimer's disease**. The particles are formed by self-assembly of a porphyrin-KLVFF peptide conjugate, in which the KLVFF sequence recognizes amyloid-beta and the porphyrin moiety acts as a light-responsive photosensitizer. Under irradiation, PKNPs produce a photothermal effect that improves blood-brain barrier penetration; interaction with amyloid-beta induces morphological disassembly and switches the construct toward photodynamic activity, enabling selective photooxygenation of amyloid-beta and inhibition of its aggregation. The system showed anti-amyloid and neuroprotective activity in preclinical Caenorhabditis elegans models, but it is not a marketed medicine or a registered clinical-stage product.
