PL-265

PL-265 is an investigational **orally active small-molecule dual enkephalinase inhibitor** developed by Pharmaleads for pain indications. It is reported to be a **prodrug of PL254** and works by inhibiting the enkephalin-degrading enzymes **neprilysin** and **aminopeptidase N**, thereby increasing local endogenous enkephalin concentrations and indirectly stimulating peripheral opioid pathways to produce analgesia. Preclinical studies have described activity in **cancer-induced bone pain**, **hyperalgesia**, and **corneal pain and inflammation**, while public reports also indicate **Phase 1 clinical evaluation in healthy volunteers** and development interest in **neuropathic chronic pain**.
