Overview

PL-4500 is a selective, orally bioavailable small molecule agonist of the melanocortin-4 receptor (MC4R), currently under development by Palatin Technologies for the treatment of obesity and metabolic disorders. The melanocortin system, particularly the MC4R subtype located in the hypothalamus, plays a critical role in the regulation of energy homeostasis, food intake, and body weight. Activation of MC4R leads to decreased appetite and increased energy expenditure. PL-4500 is part of Palatin's next-generation program aimed at developing oral small molecules that offer improved selectivity for MC4R over other melanocortin receptor subtypes (such as MC1R and MC3R), thereby potentially reducing the risk of side effects like skin hyperpigmentation or cardiovascular complications (e.g., hypertension) associated with non-selective agonists. The compound is designed to provide a more convenient oral alternative to injectable peptide-based melanocortin therapies.
