PL-8905 is a novel, selective, synthetic cyclic peptide that acts as a potent agonist of the melanocortin receptor 4 (MC4R)[8][7][1]. It demonstrates over 100-fold selectivity for MC4R compared to MC1R and exhibits significant chemical and metabolic stability, with minimal side effects on blood pressure in preclinical studies[10][2]. Developed by Palatin Technologies for the treatment of obesity and related metabolic disorders, PL-8905 decreases food intake and body weight in preclinical animal models, with additional benefits for glucose control. Combination studies with GLP-1 agonists indicate the potential for synergistic effects, yielding more significant weight loss and glucose reduction than either monotherapy. As of late 2024, PL-8905 remains in preclinical development with announced plans for IND-enabling activities and a planned phase 1 trial for obesity[8][7][1][3][5][10].
