PL-9610 is an orally bioavailable, highly selective small molecule agonist of the melanocortin 4 receptor (MC4R), currently under development by Palatin Technologies. The melanocortin system, specifically the MC4R pathway in the hypothalamus, is a critical regulator of energy homeostasis, appetite, and body weight. PL-9610 is designed to activate this pathway to reduce food intake and increase energy expenditure, offering a potential therapeutic approach for rare genetic obesity disorders. These disorders, which include deficiencies in the leptin-melanocortin signaling pathway such as POMC or LEPR deficiency, often result in early-onset, severe obesity that is refractory to standard weight-loss interventions. As a next-generation candidate, PL-9610 aims to provide the efficacy of MC4R activation with the convenience of oral administration, distinguishing it from earlier injectable peptide-based therapies.
