The PL1 aptamer is a single-stranded DNA oligonucleotide designed to specifically bind to Programmed Death-Ligand 1 (PD-L1). By antagonizing the interaction between PD-1 on T cells and PD-L1 on tumor cells, PL1 aims to restore T cell function and inhibit tumor growth. This mechanism is crucial in cancer immunotherapy, where blocking the PD-1/PD-L1 pathway prevents cancer cells from evading immune surveillance. The aptamer has shown similar tumor inhibition efficacy and binding capacity to antibodies, offering a potential alternative therapeutic agent due to its smaller size and potentially lower cost. Preclinical studies have investigated its effect on CT26 colon carcinoma.
