Overview

PL7737 is a highly selective, orally active small molecule agonist of the melanocortin-4 receptor (MC4R), developed by Palatin Technologies for the treatment of rare genetic obesity disorders. Its primary indication is obesity due to leptin receptor (LEPR) deficiency, a condition characterized by severe early-onset obesity and constant hunger from a young age. PL7737 has received FDA orphan drug designation for this indication. Preclinical studies have shown that oral administration of PL7737 significantly decreases food intake and body weight in diet-induced obese mice via MC4R-mediated mechanisms, with no effect in MC4R knockout mice—validating its target specificity. The drug lacks erectogenic activity, which is considered beneficial compared to other MC4R agonists. Clinical trials are planned to begin after an IND submission in late 2025, with additional exploration for hypothalamic obesity also underway[1][2][3][6].
