Overview

PL8177 is a synthetic cyclic heptapeptide and a potent, selective agonist of the human melanocortin-1 receptor (MC1R), with sub-nanomolar affinity binding and EC50 functional values. It is being developed primarily for the treatment of inflammatory diseases, especially ulcerative colitis and other inflammatory bowel diseases. The drug acts locally in the colon by activating MC1R on epithelial cells and resident macrophages to reduce inflammation without systemic absorption, potentially offering efficacy with fewer safety concerns compared to traditional immunosuppressive or steroid therapies. Developed by Palatin Technologies, oral formulations of PL8177 are protected from degradation in the upper GI tract and deliver the active compound directly to the colon[1][2][3][4][5][6][8].
