PL9588 is a synthetic peptide and novel melanocortin receptor agonist developed as a topical therapy for glaucoma. It activates melanocortin receptors 1 and 5 (MC1R and MC5R), resulting in a dual mechanism of action: lowering intraocular pressure (IOP) by increasing aqueous outflow through the trabecular meshwork, and providing direct neuroprotection to retinal neurons independent of IOP reduction. Preclinical studies have demonstrated that PL9588 reduces IOP comparably to standard treatments like latanoprost, with effects lasting up to 24 hours after administration. Additionally, it significantly decreases retinal cell stress/death (~25% reduction) and protects retinal ganglion cells in models of ischemia/reperfusion injury. This dual-action profile addresses an unmet need in glaucoma therapy by targeting both pressure-related damage and neurodegeneration[2][3][6][7].
