PL9643 is a synthetic peptide and melanocortin receptor pan-agonist (not active at the melanocortin 2 receptor) developed for the treatment of inflammatory ocular diseases, primarily dry eye disease (DED). It acts as an agonist at melanocortin type 1 and type 5 receptors, modulating inflammation by promoting tissue healing processes and maintaining immunologic homeostasis. PL9643 is administered as an ophthalmic solution. Clinical trials have demonstrated that it provides statistically significant improvements in both signs and symptoms of DED with a favorable safety profile. If approved, it would represent a first-in-class therapy for DED.
