PLA-1 + cisplatin is an experimental nanoparticle-based drug delivery system designed for the treatment of hepatocellular carcinoma (HCC). It consists of a metal-organic framework (MOF), designated as "1", synthesized from an organic ligand (L) coordinated with copper (Cu(II)) and iodine (I(I)), which is then incorporated with polylactic acid (PLA) to form a composite carrier (PLA-1). This carrier is loaded with cisplatin, a platinum-based chemotherapeutic agent. The system is designed to provide controlled, sustained release of cisplatin, thereby enhancing its efficacy and reducing systemic side effects. In preclinical studies using HepG-2 and Huh-7 cell lines, the formulation demonstrated dose-dependent cytotoxicity, significantly upregulating the expression of the tumor suppressor GADD45A and downregulating the oncogene NACC1.
