Overview

PLA2R-CAART is a preclinical, autologous chimeric autoantibody receptor T cell therapy designed for the treatment of PLA2R-associated membranous nephropathy, a B cell-mediated autoimmune kidney disease. The therapy comprises patient-derived T cells engineered to express a chimeric receptor that incorporates immunodominant epitopes from the human phospholipase A2 receptor (PLA2R) targeted by autoantibodies in patients with this disease. These CAAR T cells are designed to specifically recognize and eliminate B cells that produce anti-PLA2R autoantibodies, sparing healthy B cells, and thus offer a precision immunotherapy platform distinct from traditional generalized immunosuppressive therapies like rituximab. Preclinical data demonstrate selective cytolysis of pathogenic B cells without detectable off-target activity. The therapy is being developed by Cabaletta Bio and remains in preclinical and early clinical development stages[1][3][5][7].
