Drug intelligence / Profile preview
PLA2R1 monoclonal antibodies are targeted immunotherapies designed to bind to phospholipase A2 receptor 1 (PLA2R1), a transmembrane receptor expressed on glomerular podocytes that serves as the primary autoantigen in membranous nephropathy (MN). In patients with PLA2R1-associated MN, circulating autoantibodies bind to PLA2R1, forming immune complexes that activate the complement system and cause podocyte injury and proteinuria. Monoclonal antibodies targeting PLA2R1 are being investigated both as research tools to study disease pathogenesis and as potential epitope-specific therapeutic agents. For instance, patient-derived anti-PLA2R1 monoclonal antibodies (such as those cloned from memory B cells of MN patients by researchers at the University Medical Center Hamburg-Eppendorf and Novartis) have been shown to competitively inhibit the binding of pathogenic patient autoantibodies to PLA2R1 and block complement activation both *in vitro* and *in vivo*.
