PLAC8 shRNA is a gene-silencing therapeutic candidate designed to target and reduce the expression of the Placenta-specific protein 8 (PLAC8) gene. In the context of prostate carcinogenesis, particularly that induced by cadmium exposure, PLAC8 is upregulated and contributes to malignant transformation by facilitating defective autophagy and activating pro-survival pathways such as NF-κB. By utilizing short hairpin RNA (shRNA) to knock down PLAC8, the treatment induces severe endoplasmic reticulum (ER) stress through the PERK/ATF4 pathway, leading to caspase-mediated apoptosis in cancerous cells. Preclinical evidence in xenograft mouse models has shown that PLAC8 inhibition significantly suppresses tumor growth and shuts down NF-κB-mediated signaling, highlighting its potential as a targeted intervention for prostate cancer.
