placebo + cisplatin + docetaxel or paclitaxel refers to a control treatment regimen used in clinical trials for advanced primary adenocarcinoma of the lung, most notably in studies sponsored by BioNumerik Pharmaceuticals. This regimen combines a placebo with a standard-of-care chemotherapy doublet consisting of the platinum-based agent cisplatin and a taxane, which can be either docetaxel or paclitaxel. Cisplatin functions by forming DNA cross-links, thereby inhibiting DNA replication and transcription, leading to apoptosis in rapidly dividing cancer cells. The taxane component (docetaxel or paclitaxel) works by binding to and stabilizing microtubules, preventing their disassembly and arresting the cell cycle in the G2/M phase. This specific combination was utilized as the comparator arm in the Phase 3 trial of Tavocept (BNP7787) to evaluate whether the addition of Tavocept could improve survival outcomes or mitigate chemotherapy-induced toxicities such as neuropathy and nephrotoxicity.
