Placebo + fluorouracil + cisplatin is a combination therapy used as the control arm in the GEMSTONE-304 Phase 3 clinical trial conducted by CStone Pharmaceuticals. This regimen consists of a placebo administered in conjunction with a standard chemotherapy doublet of fluorouracil and cisplatin (FP). Fluorouracil is an antimetabolite that inhibits thymidylate synthase, while cisplatin is a platinum-based agent that causes DNA cross-linking. The combination was evaluated as a first-line treatment for patients with unresectable locally advanced, recurrent, or metastatic esophageal squamous cell carcinoma (ESCC) to serve as a comparator for the PD-L1 inhibitor sugemalimab.
