This combination refers to the control arm of clinical trials conducted by OPKO Health (specifically through its subsidiary Transition Therapeutics) for the treatment of type 2 diabetes mellitus. In these studies, patients receive a placebo (administered as a daily subcutaneous injection) in addition to their existing background standard-of-care therapies, which include the biguanide metformin and/or insulin-sensitizing thiazolidinediones (TZDs). Metformin acts primarily by inhibiting hepatic gluconeogenesis and improving peripheral insulin sensitivity via the activation of AMP-activated protein kinase (AMPK). Thiazolidinediones are agonists of the peroxisome proliferator-activated receptor gamma (PPAR-gamma), which regulates the transcription of genes involved in glucose and lipid metabolism. This specific combination was used as the comparator for the gastrin-17 analogue TT223 in Phase 2 clinical evaluations.
